Poster: Faster Generation of Anti-Drug Antibodies Using SpyTag Technology

This poster shows how the Bio-Rad custom antibody team generated anti-idiotypic antibodies using antibody phage display and in vitro guided selection. Through the incorporation of SpyTag into recombinant antibodies, we show how a monovalent Fab fragment can be converted to an IgG or a bivalent Fab in just one hour.

Furthermore, SpyTag technology offers fast and robust site-specific labeling of antibodies using preproduced conjugated SpyCatcher modules. We demonstrate how a more sensitive pharmacokinetic bridging ELISA can be developed with these antibodies, compared to using a conventionally labeled alternative.

An inhibitory anti-idiotypic antibody can be used as a surrogate positive control or calibrator in an anti-drug antibody bridging ELISA. Learn how using SpyTag technology enables multiple antibodies to be tested in an IgG-like format, offering a cost-effective way to select the most promising candidates to convert to the fully human format for the final clinical assay.
