This poster shows how the Bio-Rad custom antibody team generated anti-idiotypic antibodies targeting a chimeric antigen receptor (CAR) using antibody phage display and in vitro guided selection.
The workflow took advantage of recombinant antibodies incorporating a SpyTag, which enabled rapid format switching and site-directed conjugation. By coupling to a set of preprepared SpyCatchers, different formats of the SpyTagged antibodies were created to determine specificity and affinity and screen in flow cytometry.
Antibodies that distinguish between an scFv-expressing natural killer (NK) cell line and negative control cells, as well as CAR T-cells and unrelated CAR T-cells, were selected.
