How to Choose a Primary Antibody for Multiplex Western Blotting

In this video, we discuss the selection of primary antibodies:

  • Choosing primary antibodies from different host species is often the easiest way to multiplex
  • When choosing antibodies from different host species, distantly related species are less likely to have issues with species cross-reactivities. Avoid choosing antibodies from closely related species like mouse and rat
  • Using primary antibodies from the species but different antibody classes like IgG and IgM in combination with appropriate secondary antibodies is one way to overcome species issues
  • Primary antibodies from the same species but of different isotypes such as IgG-1 and IgG-2 provide another possible solution

How to Choose Secondary Antibodies for Multiplex Western Blotting

In this video, we discuss the best practices for selecting secondary antibodies:

  • Use the correct secondary antibodies to detect your primary antibodies. This will be a species-specific, class-specific, or isotype-specific approach depending on your multiplexing approach
  • Do not use secondary antibodies raised in the same species as any of your primary antibodies when using a species-specific approach
  • Ensure specificity of your staining by using cross-adsorbed secondary antibodies to prevent cross-reaction with primary antibodies. The cross-adsorbed antibody will correctly bind the target primary antibody but will not cross-react to the primary antibody from the related species
  • Ensure that the different detection methods can be detected by your imager
