Human alpha fetoprotein (PHP152) detected in a sandwich ELISA using Mouse anti Human alpha fetoprotein (MCA5862G) as the capture reagent and HRP-conjugated Mouse anti Human alpha fetoprotein (MCA5861G) as the detection reagent. HRP-conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit®

Sandwich ELISA analysis of alpha fetoprotein binding using Mouse anti Human alpha fetoprotein (4520-5306) as a capture reagent and biotinylated Mouse anti Human alpha fetoprotein (MCA5862) as a detection reagent with human alpha fetoprotein (Bio-Rad Catalog 13752600) as antigen for the generation of a standard curve. Detection is by HRP conjugated Streptavidin and substrate. Microtitre plate is read at O.D. 450 nm on the Bio-Rad iMark Microplate Absorbance Reader (Bio-Rad Catalog 168-1135). Serum (green) and plasma (orange) samples are both displayed at 1:2 dilution.

    • Mouse anti Human alpha fetoprotein (clone 125.6G7) is a monoclonal antibody recognising alpha fetoprotein (AFP), a member of the albuminoid protein superfamily, along with albumin and vitamin D binding protein. Members of the albuminoid superfamily comprise molecules with three characteristic globular domains (Mizejewski 1995) .

AFP is a ~70kD plasma protein found in fetuses over four weeks old, produced by the yolk sac and the fetal liver, with the highest concentration reached during the 12th to 16th week of gestation. AFP acts as a transport protein for various ligands such as bilirubin, fatty acids, steroids and heavy metals. After birth, plasma AFP levels drop rapidly with only trace amounts remaining in healthy adults. Changes in plasma levels of AFP have been documented in a variety of cancers including hepatocellular cancer (Sato et al. 1993)

Clone 125.6G7 shows <0.1% reacativity with human serum albumin and IgG. Strong expression of AFP has been noted in the hepatic cell line HepG2.
