| Mouse anti Human Interleukin-6 antibody, clone rC01/1E8, is a recombinant monoclonal antibody that recognizes Interleukin-6 (IL-6), also known as B cell stimulatory factor 2 (BSF2). IL-6 is a multifunctional cytokine composed of four alpha helices, containing various glycosylation sites and two disulfide bonds. It is expressed by numerous cell types including B lymphocytes, T lymphocytes, macrophages and dendritic cells. IL-6 secretion is stimulated during inflammation in response to tissue injury or infection. IL-6 signaling involves binding of IL-6 to either the membrane bound (classical pathway) or soluble (trans-signaling pathway) IL-6 receptor (IL-6R, CD126). IL-6/IL-6R further forms a complex with glycoprotein 130 (CD130), activating signal transduction, for example by the JAK/STAT3 pathway. IL-6 has diverse roles, including promoting B cell differentiation into plasma cells and stimulating IgG production, triggering the production of acute phase reactants by the liver, controlling lymphocyte trafficking and regulating differentiation of monocytes into macrophages. IL-6 dysregulation is involved in the pathogenesis of several autoimmune diseases. Therapeutic modulation of IL-6 has resulted in the approval of a number of treatments for autoimmune disorders such as rheumatoid arthritis, juvenile idiopathic arthritis and cytokine release syndrome. |
