| Mouse anti Human FoxP3 antibody, clone rA16B16-01/2F5, is a recombinant monoclonal antibody that recognizes FoxP3, also known as Scurfin or IPEX. FoxP3 is a member of the forkhead transcription factor family. It is believed to be a master regulatory gene and is crucial for the development and inhibitory function of regulatory T-cells (Tregs). FoxP3 plays a central role in immune system homeostasis by driving the suppressive function and stability of the Treg lineage, through which it directly modulates the function and expansion of all T cells. The suppressive activity of Tregs involves the coordinated activation of several genes by FoxP3, including CTLA-4 and TNFRSF1, along with the repression of genes encoding inflammatory cytokines, such as interleukin-2 (IL-2) and interferon-gamma (IFNγ). FoxP3 is downregulated during inflammation to allow for the acquisition of effector T cell-like functions. FoxP3 inhibits cytokine production and T-cell effector function by repressing the activity of two key transcription factors, RELA and NFATC2. FoxP3 is a key marker for CD4+ Tregs. FoxP3-expressing CD4+ Tregs have a key role in immune tolerance and suppression of autoimmune and anti tumor immune responses. FoxP3 deficiency causes immune dysregulation and enteropathy X-linked syndrome (IPEX). |
