| Mouse anti Human CD85k antibody, clone rA43B43-02/1F6-3, is a recombinant monoclonal antibody that recognizes CD85k, also known as Immunoglobulin-like transcript 3 (ILT-3) and leukocyte immunoglobulin-like receptor subfamily B member 4 (LILRB4). CD85k is a type I transmembrane glycoprotein, part of the LILR family of innate immunomodulatory receptors. It contains two extracellular immunoglobulin-like domains and three cytoplasmic immunoreceptor tyrosine-based inhibitor motifs (ITIM) that exert an inhibitory effect on immune responses. CD85k is expressed by dendritic cells, monocytes and macrophages and is stimulated by a range of ligands, including APOE, CD166 and fibronectin. Upon ligand binding, the Src-family protein Lyn is autophosphorylated, phosphorylating the ITIM tyrosine residues and leading to negative regulation of Syk and PI3K signaling. CD85k has roles in the induction of tolerogenic dendritic cells and regulatory T cells, inhibition of cytokine production and inhibition of T cell activity. CD85k is expressed on a number of cancers, including multiple myeloma, gastric cancer, AML and ovarian cancer. It is a potential therapeutic target in cancer immunotherapy. |
