| Mouse anti Human CD73 antibody, clone rA9-03/4C2, is a recombinant monoclonal antibody that recognizes CD73, also known as Ecto-5'-nucleotidase or 5’-NT. CD73 is a cell surface GPI-anchored enzyme that catalyses the final step in the extracellular metabolism of ATP to form adenosine. It is expressed on endothelial cells, subsets of T cells and B cells, follicular dendritic cells and stromal cells. The formation of extracellular adenosine by CD73 regulates numerous signaling pathways through the activation of G protein-coupled adenosine receptors. CD73 plays a critical role in cellular homeostasis, stress responses and injury across many tissues. CD73 was originally identified as a marker of lymphocyte differentiation. CD73 is expressed by many different types of cancer and promotes tumor outgrowth, metastasis and drug resistance. High CD73 expression in tumors is associated with shorter overall survival and poor prognosis of patients in multiple cancers, including ovarian cancer, pancreatic cancer, and melanoma. Up-regulation of CD73 in breast cancer patients has been highlighted as a mechanism of resistance to therapies, with CD73 expression significantly related to poor prognosis and chemoresistance and blockade of the enzyme improving antitumor responses. CD73 expression has been indicated as both a potential prognostic factor for human solid tumors, a promising target in future cancer immunotherapy and as a potential biomarker for anti-PD-1/PD-L1 treatment. |
