| Mouse anti Human CD46, clone rA44B44-08/4D7, is a recombinant monoclonal antibody that recognizes CD46, also known as membrane cofactor protein (MCP), Trophoblast leukocyte common antigen or TLX. CD46 is a single-pass type I transmembrane protein expressed by almost all nucleated cells. Four major isoforms exist, each of which has an amino terminus containing four complement control protein modules and an alternatively spliced O-glycosylated domain. The cytoplasmic region consists of an anchor region and an alternatively spliced cytoplasmic tail containing a distinct signaling motif. CD46 has a role in regulating complement activation by binding C3b and C4b after deposition on a host cell, facilitating cleavage. The CD46 C2 isoform is exclusively expressed on the inner acrosomal membrane of human spermatozoa, where is serves a role in fertilization. CD46 is also involved in T cell activation, autophagy and the intracellular complement system. CD46 is often dysregulated in cancer, for example in multiple myeloma and various solid tumors. It has been reported as a potential tumor target and prognostic indicator. CD46 is a receptor or adherence factor for a number of pathogens, including the measles virus. |
