| Mouse anti Human CD40 antibody, clone rA38B38-06/3H2, is a recombinant monoclonal antibody that recognizes CD40, also known as Bp50 or Tumor necrosis factor receptor superfamily member 5. CD40 is a type 1 transmembrane protein belonging to the Tumor Necrosis Factor Receptor (TNFR) superfamily. It is expressed on a variety of immune cells, including B lymphocytes, dendritic cells (DCs) and monocytes. The ligand for CD40 (CD154) is predominantly expressed on the surface of activated T cells. Ligand binding to CD40 activates a range of downstream signaling pathways including the canonical and non-canonical NF-κB, PI3 kinase and MAP kinase pathways. On DCs, CD40 engagement by CD40 ligand expressing CD4+ T cells stimulates the production of proinflammatory cytokines and increases the expression of co-stimulatory molecules such as CD80. DCs are licensed to present exogenous molecules to CD8+ T cells, resulting in their activation. On B cells, CD40 engagement results in B cell activation, proliferation and differentiation. CD40 can be used as a surface marker for B cells. It is a potential therapeutic target for treatment of cancer. |
