| Mouse anti Human CD39 antibody, clone rA8-07/4C11, is a recombinant monoclonal antibody that recognizes CD39, also known as Ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1) or NTPDase1. CD39 is an integral membrane protein that dephosphorylates extracellular ATP (eATP) to AMP. CD39 is the rate limiting enzyme in the generation of extracellular adenosine. Accumulated extracellular adenosine mediates broad immunosuppressive actions by binding adenosine receptors such as A2AR. CD39 is expressed on several immune subpopulations, particularly B cells, natural killer (NK) cells, tumor-associated-macrophages (TAM) and tumor-infiltrating lymphocytes (TILs) including Tregs. It is also highly expressed in placenta, lung, skeletal muscle and kidney. CD39 can be used as a marker to distinguish Tregs from other resting or activated T cells. Overexpression of CD39 has been found in several cancers, including ovarian cancer, breast cancer, lymphoma, colon cancer and melanoma, with high expression levels considered an indicator of poor disease outcomes and disease progression. The upregulation of CD39 by tumors is therefore implicated in depleting the immune-stimulatory eATP in the tumor microenvironment (TME), leading to tumor immune evasion. |
