Mouse anti Human CD163 antibody, clone rAB04, is a recombinant monoclonal antibody that recognizes CD163, also known as hemoglobin scavenger receptor or scavenger receptor cysteine-rich type 1 protein M130.

CD163 is a high affinity scavenger receptor for the haptoglobin-hemoglobin (Hp-Hb) complex, expressed by tissue resident macrophages and circulating blood monocytes. It consists of a single transmembrane domain, a short cytoplasmic tail and a large extracellular region containing nine scavenger receptor cysteine-rich (SRCR) domains. CD163 is involved in the removal of the Hp-Hb complex through macrophage endocytosis and therefore aids in protection from free hemoglobin-mediated oxidative damage. A soluble form of CD163 is generated by enzymatic cleavage of membrane-bound CD163 and is thought to have an anti-inflammatory role.

CD163 is considered a classical M2 macrophage marker. It is upregulated when macrophages switch to alternative activated phenotypes in inflammation. The soluble form of CD163 is being investigated as a diagnostic tool for macrophage activation in inflammatory conditions such as atherosclerosis.

This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.

Avoid repeated freezing and thawing as this may denature the antibody. Storage in frost-free freezers is not recommended.

Flow Cytometry: Use 5μl of the suggested working dilution to label 0.5x10⁶ cells in 100μl. Best practices suggest a 5 min centrifugation at 6,000g prior to sample application.
