Mouse anti Human CD127 antibody, clone rAbD11590kb, is a recombinant monoclonal antibody that recognizes CD127, also known as human interleukin-7 receptor subunit alpha (IL-7Rα).

CD127 is a single-pass type I transmembrane protein that associates with CD132, the common cytokine receptor γ-chain, to form the functional high affinity IL-7 receptor. CD127 expressing cells include early thymocytes, T cells, pre-B cells and bone marrow macrophages. IL-7 mediated signaling through IL-7R activates two major pathways, the JAK/STAT5 and Akt/PI3K pathways, and plays a critical role in immune cell development. IL-7R deficiency severely impairs the development of immune cells.

Alongside other surface antigens, CD127 can be used as a marker for regulatory T cells. IL-7 signaling through CD127 is impaired in cancers and chronic infections.

This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.

Avoid repeated freezing and thawing as this may denature the antibody. Storage in frost-free freezers is not recommended.

Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.

Flow Cytometry: Use 5μl of the suggested working dilution to label 0.5x10⁶ cells in 100μl. Best practices suggest a 5 min centrifugation at 6,000g prior to sample application.
