Mouse anti Human C5a/C5a desArg (neo-epitope) antibody, clone 2952, recognizes a neo-epitope on the human complement proteins C5a and C5a desArg. Neo-epitopes are regions on native proteins, like C5, that are normally only exposed and detectable by monoclonal antibodies, on active forms of the protein. C5 is a member of the complement cascade and all three activation pathways (classical, lectin and alternative), lead to its cleavage. Upon activation of any of these pathways, C5 is cleaved by C5 convertases into C5b, which mediates pathogen lysis, and the inflammatory mediator C5a. The inflammatory responses mediated by C5a are chemotaxis, leukocyte degranulation, vascular permeability and cytokine plus chemokine production. Beyond innate immunity though, C5a plays a role in organ development, tissue regeneration and hematopoiesis (Zhang et al. 2017). C5a elicits these responses through its interaction with the receptors C5aR and C5aR (C5L2). These receptors are expressed ubiquitously, but are found in higher levels on the surface of immune cells, including macrophages, neutrophils and T cells. However, C5a is a short lived in circulation and is cleaved at its C-terminal arginine, by carboxypeptidases, generating the more stable C5a desArg. While C5a desArg is more stable in serum than C5a, it is less potent in comparison, but they do activate a similar range of biological responses (Zhang et al. 2017, Merle et al. 2015). |
