| Mouse anti Human C5/C5a antibody, clone 557, recognizes an epitope on C5 and C5a. C5 is a component of all three complement activation pathways (classical, lectin and alternative), which lead to its cleavage by C5 convertase. Cleavage of C5 results in the formation of C5b, which initiates pathogenic lysis and C5a, a potent inflammatory mediator. C5a is a ligand for the G protein coupled receptors, C5aR1 and C5aR2 (also known as C5L2) through which its inflammatory functions are controlled (Merle et al. 2015). These receptors are expressed ubiquitously but are found at its highest levels on the surface of immune cells, including macrophages, neutrophils and T cells. The inflammatory responses mediated by C5a are chemotaxis, leukocyte degranulation, vascular permeability and cytokine plus chemokine production. Beyond innate immunity, C5a plays a role in organ development, tissue regeneration and hematopoiesis (Zhang et al. 2017). |
