Infliximab antibody | AbD20436
A microtiter plate was coated over night with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST+5% BSA, detection was performed using Human anti infliximab (clone 20436) at a concentration of 2 μg/ml followed by an HRP-labelled anti-His tag in HISPEC assay diluent (BUF049A) and a fluorogenic peroxidase substrate.
A microtiter plate was coated over night with human IgG1/kappa, human IgG1/lambda, cetuximab or infliximab at a concentration of 5μg/ml. After washing and blocking with PBST+5% BSA, detection was performed using Human anti infliximab (clone AbD20436) titrated to the given concentrations in PBST+10% human serum, followed by an HRP-labelled anti-His tag in HISPEC assay diluent (BUF049A) and a fluorogenic peroxidase substrate.

Detection was performed using HRP-conjugated Human anti infliximab, clone 19370_hIgG1, (HCA215) in HISPEC assay diluent (BUF049A) and a fluorogenic peroxidase substrate. HRP-conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK001P).

After washing and blocking with PBST+5% BSA, 10% human serum was added spiked with increasing concentrations of infliximab. Detection was performed using HRP-conjugated Human anti infliximab, clone AbD20436_hIgG1 (HCA233) in HISPEC assay diluent (BUF049A) and a fluorogenic peroxidase substrate. HRP-conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK001P).
A microtiter plate was coated over night with TNFα at a concentration of 1 μg/ml. After washing and blocking with PBST+5% BSA, a pre-incubated mixture of infliximab (IFX, 0.3 μg/ml) plus increasing concentrations of Human anti infliximab (clone AbD20436) was added. Free infliximab, still capable of binding to the TNFα-coated plate, was detected using Goat anti Human IgG F(ab')2:HRP (0500-0099).

| Human anti Infliximab, clone AbD20436, is an affinity matured anti-idiotypic antibody in monovalent Fab format that specifically recognizes free infliximab but not infliximab bound to its target, Tumor Necrosis Factor Alpha (TNFα). It can be used in bioanalytical assays to measure the levels of infliximab and biosimilar products, such as Inflectra and Renflexis, in patient samples. Human anti Infliximab, clone AbD20436 inhibits the binding of infliximab to TNFα and can be used as a capture antibody in a pharmacokinetic (PK) assay in the bridging format to measure free drug. Infliximab (branded as Remicade®) is a chimeric IgG1 kappa monoclonal antibody approved for the treatment of psoriasis, Crohn's disease, ankylosing spondylitis, psoriatic arthritis, rheumatoid arthritis and ulcerative colitis. Infliximab is directed against TNFα and acts by blocking the binding of TNFα to its receptors, resulting in a down-regulation of the inflammatory response associated with autoimmune diseases.
