Western blot analysis of RelB was performed by loading 20 ug of THP-1 (lane1), Daudi (lane2) and Raji (lane3) cell lysate on a gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5 % skimmed milk for 1 hour at room temperature. RelB was detected at ~ 63 kDa using Mouse anti RelB (MCA6037) at 1-2 ug/ml in 5 % skimmed milk at 4°C overnight on a rocking platform. Immunofluorescence analysis of RelB was performed on 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.25% Triton™ X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. The cells were labeled with Mouse anti RelB antibody (MCA6037) at 1.0 ug/ml in 1% BSA and incubated for 3 hours at room temperature and then labeled with an Alexa Fluor® 488 conjugated goat anti-mouse IgG secondary antibody for 45 minutes at room temperature (Panel A: green). Nuclei (Panel B: blue) were counterstained with DAPI and F-actin (Panel C: red) with rhodamine phalloidin. Panel D is a merged image showing nuclear localization. Panel E is a no primary antibody control. The images were captured at 60x magnification

    • Mouse anti-RelB antibody recognizes the transcription factor RelB, also known as I-Rel. RelB is encoded by the v-rel avian reticuloendotheliosis viral oncogene homolog B gene and is a critical component of the NF-kappa-B pathway.

      Upon cleavage of the NF-kappa-B p105 and p100 subunits into the NF-kappa-B p50 and p52 subunits, RelB forms RelB-p50 and RelB-p52 heterodimers. These dimers act as activators of transcription by binding to kappaB sites present on DNA (Oeckinghaus and Ghosh 2009). RelB-p50 and RelB-p52 binding induces the expression of genes mediating immune responses, inflammation and apoptosis (Jing and Lee 2014).

      Misregulation of the NF-kappa-B pathway has been implicated in a variety of disease states including autoimmunity and cancer. As the NF-kappa-B pathway has been demonstrated to be constitutively active in a large group of tumors, developing NF-kappa-B inhibitors has emerged as a new cancer therapy approach (Chaturverdi et al. 2011, Baud and Karin et al. 2009).
