Rat anti Mouse TREM-2 antibody, clone 78.18 (MCA4772EL) used as a blocking reagent on primary microglia to look at the interaction between TREM2 and Aβ monomers.

i) Primary microglia were incubated with 100 nM HiLyte Fluor 647-labelled Aβ42 at 4°C for 1 hour, then lysed, immunoprecipitated with anti-TREM2 antibodies, and examined by SDS-PAGE and western blotting. Aβ oligomers co-immunoprecipitate with endogenous TREM2 in primary microglia (lane 2), but not with other natively endogenous Type I glycoproteins, such as nicastrin (lane 4) or with isotype control antibody (lane 3). TREM2 knockout microglia failed to co-immunoprecipitate Aβ (lane 1). ii) This interaction was blocked by preincubation with monoclonal anti-TREM2 blocking antibody (lane 2), but not by isotype control antibodies (lane 3).

F BA Reset| Rat anti Mouse TREM-2 antibody, clone 78.18 recognizes triggering receptor expressed on myeloid cells 2 (TREM-2), an activating receptor of the Ig superfamily. TREM-2 forms a receptor signaling complex with DAP12 to trigger the activation of the immune response in macrophages and dendritic cells. It may be involved in chronic inflammation and may promote the production of constitutive rather than inflammatory chemokines and cytokines. TREM-2 may also have a functional role in osteoclast formation. Stimulation of TREM-2 with Rat anti Mouse Trem-2 antibody, clone 78.18 enhances multinucleated osteoclast formation in vitro. Rat anti Mouse Trem-2 antibody, clone 78.18 may also be used to block TREM-2 on pre-osteoclasts to inhibit the formation of multinuclear osteoclasts and to inhibit bone resorption by mature osteoclasts. |
