Pertuzumab antibody | AbD37070
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Pertuzumab Antibody, clone AbD37070 (HCA342) at a concentration of 2 μg/ml followed by an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with human HER2, pertuzumab, trastuzumab, human IgG1 kappa or lambda at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the human HER2/pertuzumab complex was formed by adding 5 µg/ml pertuzumab to the wells coated with human HER2. Detection was performed using Human Anti-Pertuzumab Antibody, clone AbD37070 (HCA342) titrated to the given concentrations in PBST followed by Mouse Anti-Penta Histidine Tag:HRP Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for Pertuzumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of pertuzumab. Detection was performed using HRP conjugated Human Anti-Pertuzumab Antibody, clone AbD37069ia (HCA345) at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of pertuzumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Pertuzumab Antibody, clone AbD37070 (HCA342) was added. Free pertuzumab, still capable of binding to the HER2 coated plate, was detected using an HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Pertuzumab Antibody, clone AbD37070 is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the humanized monoclonal antibody pertuzumab. It does not recognize free human epidermal growth factor receptor 2 (HER2) nor pertuzumab in complex with HER2. The antibody can be used to measure free pertuzumab or its biosimilars in patient samples. Clone AbD37070 is a fully human recombinant monoclonal antibody in monovalent Fab format and is suitable as a capture antibody for pertuzumab in a pharmacokinetic (PK) bridging ELISA to measure free drug when paired with clone AbD37069ia (HCA345, hIgG1 format) as the detection reagent. Pertuzumab (Perjeta) is an IgG1/kappa antibody humanized from mouse. It has been approved for the treatment of patients with metastatic HER2-positive breast cancer in combination with trastuzumab. Pertuzumab binds to the extracellular domain II of HER2 and inhibits HER2–HER3 dimerization, reducing signaling through intracellular pathways (Capelan et al. 2013). This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.
