Evolocumab antibody | AbD40410ia
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Evolocumab Antibody, clone AbD40410ia (HCA376) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Evolocumab Antibody, clone AbD40410ia (HCA376) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Detection was performed using HRP conjugated Human Anti-Evolocumab Antibody, clone AbD40410ia (HCA376) at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of evolocumab (0.3 µg/ml) plus increasing concentrations of Human Anti-Evolocumab Antibody, clone AbD40410 in the monovalent Fab format was added. Free evolocumab, still capable of binding to the human PCSK9 coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. | Human Anti-Evolocumab Antibody, clone AbD40410ia, is a paratope specific, inhibitory antibody that specifically recognizes the monoclonal antibody drug evolocumab. It does not recognize the drug target, human proprotein convertase subtilisin/kexin type 9 (PCSK9), nor evolocumab in complex with PCSK9. The antibody can be used to measure free evolocumab in patient samples. A pair of anti-evolocumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. This antibody can be used as the detection antibody, paired with an anti-evolocumab antibody in monovalent Fab format, clone AbD40406ad (TZA007), as the capture antibody. Clone AbD40410ia is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 5 nM and is one of three antibodies offered for ADA assays. Antibody HCA377 has affinity 1 nM and HCA378 has affinity 5 nM. Evolocumab (Repatha) is a fully human IgG2 lambda antibody developed using transgenic mouse technology. It has been approved for treating selected patients with high cholesterol, and for the prevention of heart attack and stroke. The drug prevents the PCSK9 protein binding to the low density lipoprotein receptor (LDLR), thereby increasing the number of LDLRs available and lowering LDL levels.
