Evolocumab antibody | AbD40406ad
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Evolocumab Antibody, clone AbD40406ad (TZA007) at a concentration of 2 μg/ml and an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with evolocumab, human PCSK9, human IgG1 kappa or human IgG1 lambda at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Evolocumab Antibody, clone AbD40406ad (TZA007) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of evolocumab (0.3 μg/ml) plus increasing concentrations of Human Anti-Evolocumab Antibody, clone AbD40406ad (TZA007) was added. Free evolocumab, still capable of binding to the human PCSK9 coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. | Human Anti-Evolocumab Antibody, clone AbD40406ad, is a paratope specific, inhibitory antibody that specifically recognizes the monoclonal antibody drug evolocumab. It does not recognize the drug target, human proprotein convertase subtilisin/kexin type 9 (PCSK9), nor evolocumab in complex with PCSK9. The antibody can be used to measure free evolocumab in patient samples. The antibody is tagged with a SpyTag2 at the C-terminus of the Fab antibody heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody, in monovalent Fab format, is recommended as the capture antibody, use the same clone in fully human IgG1 format, clone AbD40410ia (HCA376) for detection. Evolocumab (Repatha) is a fully human IgG2 lambda antibody developed using transgenic mouse technology. It has been approved for treating selected patients with high cholesterol, and for the prevention of heart attack and stroke. The drug prevents the PCSK9 protein binding to the low density lipoprotein receptor (LDLR), thereby increasing the number of LDLRs available and lowering LDL levels.
