Evolocumab antibody | AbD40410
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Evolocumab Antibody, clone AbD40410pap (TZA008P) at a concentration of 0.2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Evolocumab Antibody, clone AbD40410pap (TZA008P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of evolocumab. Detection was performed using HRP conjugated Human Anti-Evolocumab Antibody, clone AbD40410pap (TZA008P) at a concentration of 0.5 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human PCSK9 at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of evolocumab (0.3 μg/ml) plus increasing concentrations of Human Anti-Evolocumab Antibody, clone AbD40410 (TZA008) was added. Free evolocumab, still capable of binding to the human PCSK9 coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. | Human Anti-Evolocumab Antibody, clone AbD40410, is a paratope specific, inhibitory antibody that specifically recognizes the monoclonal antibody drug evolocumab. It does not recognize the drug target, human proprotein convertase subtilisin/kexin type 9 (PCSK9), nor evolocumab in complex with PCSK9. The antibody can be used to measure free evolocumab in patient samples. The monovalent Fab format of this antibody, clone AbD40410ad (TZA008) is tagged with a SpyTag2 at the C-terminus of the Fab antibody heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is recommended as the detection antibody when in the bivalent HRP conjugated format, clone AbD40410pap (TZA008P) paired with an anti-evolocumab antibody in monovalent Fab format, clone AbD40406ad (TZA007), as the capture antibody. Evolocumab (Repatha) is a fully human IgG2 lambda antibody developed using transgenic mouse technology. It has been approved for treating selected patients with high cholesterol, and for the prevention of heart attack and stroke. The drug prevents the PCSK9 protein binding to the low density lipoprotein receptor (LDLR), thereby increasing the number of LDLRs available and lowering LDL levels. - A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA008 with a HRP conjugated BiCatcher version 2. The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
