A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Etanercept Antibody, clone AbD26183pap (TZA0145P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with Etanercept, human TNFR2, human Fc control and Infliximab at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Etanercept Antibody, clone AbD26183pap (TZA0145P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for etanercept are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of etanercept. Detection was performed using HRP-conjugated Human Anti-Etanercept Antibody, clone AbD26183pap (TZA0145P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with human TNF-α at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of etanercept (0.3 µg/mL) plus increasing concentrations of Human Anti-Etanercept Antibody, clone AbD26183pap (TZA0145P) and AbD25940 (HCA277) in the monovalent Fab format was added. Free etanercept, still capable of binding to the human TNF-α coated plate, was detected using HRP conjugated Mouse anti Human IgG (Fc) CH2 domain antibody (MCA647P) followed by QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Etanercept Antibody, clone AbD26183pap is a non-inhibitory anti-idiotypic antibody (type 2) that specifically recognizes free humanized monoclonal antibody drug etanercept. It does not recognize free TNF-α and TNF-β but Etanercept in complex with TNF-α. This antibody can be used to measure free and bound Etanercept in serum of patients. TZA0145P, clone AbD26183pap is a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA. A pair of anti-etanercept antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. Clone AbD26183pap is recommended as the detection antibody, paired with Human Anti-Etanercept Antibody in monovalent Fab format, clone AbD25939 (HCA276) as the capture antibody. Etanercept (Enbrel®) is a dimeric fusion protein consisting of the extracellular ligand-binding portion of the human 75 kDa (p75) TNFR2 linked to the Fc portion of human IgG1. The Fc component of etanercept contains the CH2 domain, the CH3 domain and hinge region, but not the CH1 domain of IgG1. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
