Cemiplimab antibody | AbD41947ad
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, Human Anti-Cemiplimab Antibody, clone AbD41947ad (TZA006) at a concentration of 2 μg/ml was added. Detection was performed using HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with cemiplimab, human IgG4 kappa (S228P) isotype control or pembrolizumab at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Cemiplimab Antibody, clone AbD41947ad (TZA006) titrated to the given concentrations in PBST, followed by Mouse Anti- Penta Histidine Tag:HRP Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with human PD-1 at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of cemiplimab (0.15 µg/ml) plus increasing concentrations of Human Anti-Cemiplimab Antibody, clone AbD41947ad (TZA006) was added. Free cemiplimab, still capable of binding to the human PD-1 coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

Detection was performed using HRP conjugated Human Anti-Cemiplimab Antibody, clone AbD41947ia (HCA368) at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

| Human Anti-Cemiplimab Antibody, clone AbD41947ad is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the fully human monoclonal antibody cemiplimab. It does not recognize free human programmed death receptor-1 (PD-1) nor cemiplimab in complex with PD-1. The antibody can be used to measure free cemiplimab in patient samples. The antibody is tagged with a SpyTag2 at the C-terminus of the Fab antibody heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. A pair of anti-cemiplimab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. This antibody can be used as a capture reagent, paired with Anti-Cemiplimab Antibody, clone AbD41947ia (HCA368) in full IgG1 format as the detection reagent.
