A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Abatacept Antibody, clone AbD37058pap (TZA0143P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with Abatacept, Human Fc control and Human IgG1 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Abatacept Antibody, clone AbD37058pap (TZA0143P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for abatacept are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of abatacept. Detection was performed using HRP-conjugated Human Anti-Abatacept Antibody, clone AbD37058pap (TZA0143P) at a concentration of 2 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human CD80-Fc at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of HRP conjugated abatacept (1 µg/mL) plus increasing concentrations of Human Anti- Antibody, clone AbD37058pap (TZA0143P) in the monovalent Fab format was added. Free HRP conjugated abatacept, still capable of binding to the human CD80-Fc coated plate, was detected using QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Abatacept Antibody, clone AbD37058pap is a paratope specific, inhibitory antibody that specifically recognizes the free fusion protein drug abatacept as well as cytotoxic T-lymphocyte antigen-4 (CTLA-4), also known as CD152. TZA0143P, clone AbD37058pap is a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA. A pair of anti-abatacept antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD37058pap is recommended as a detection antibody, paired with Human Anti-Abatacept Antibody, clone BNI3 (MCA1724) as the capture antibody. Abatacept (Orencia) is a recombinant fusion protein consisting of the extra-cellular domain of human CTLA-4, fused to the modified Fc (hinge, CH2, and CH3 domains) portion of human IgG1. It is used to treat several autoimmune diseases, including rheumatoid arthritis, psoriatic arthritis and juvenile idiopathic arthritis, by inhibiting optimal T-cell activation. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
