A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Secukinumab Antibody, clone AbD37054ia (HCA371) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of secukinumab (0.3 µg/ml) plus increasing concentrations of Human Anti-Secukinumab Antibody, clone AbD37054ia (HCA371) in the monovalent Fab format was added. Free secukinumab, still capable of binding to the human IL-17A coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. | Human Anti-Secukinumab Antibody, clone AbD37054ia is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the human monoclonal antibody secukinumab. It does not recognize free human interleukin 17A (IL-17A) nor secukinumab in complex with human IL-17A. The antibody can be used to measure secukinumab or its biosimilars in patient samples. A pair of anti-secukinumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. Either this antibody, or clone AbD37091ia (HCA372), is recommended as the detection antibody, paired with an anti-secukinumab antibody in monovalent Fab format, clone AbD37052 (HCA370), as the capture antibody. Clone AbD37054ia is a fully human recombinant monoclonal antibody with IgG1 isotype and can be used as a surrogate positive control or reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 3 nM, and is one of three antibodies with different affinities offered for ADA assays. Secukinumab (Cosentyx) is a human IgG1/kappa antibody generated using transgenic mouse technology. It has been approved for the treatment of plaque psoriasis, ankylosing spondylitis, psoriatic arthritis and non-radiographic axial spondyloarthritis.
