Secukinumab antibody | AbD37052
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Secukinumab Antibody, clone AbD37052 (HCA370) at a concentration of 2 μg/ml followed by an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.

After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Secukinumab Antibody, clone AbD37052 (HCA370) titrated to the given concentrations in PBST followed by Mouse Anti-Penta Histidine Tag:HRP Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of secukinumab (0.3 µg/ml) plus increasing concentrations of Human Anti-Secukinumab Antibody, clone AbD37052 (HCA370) in the monovalent Fab format was added. Free secukinumab, still capable of binding to the human IL-17A coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Secukinumab Antibody, clone AbD37052 is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the human monoclonal antibody secukinumab. It does not recognize free human interleukin 17A (IL-17A) nor secukinumab in complex with human IL-17A. The antibody can be used to measure secukinumab or its biosimilars in patient samples. Clone AbD37052 is a fully human recombinant monoclonal antibody in monovalent Fab format and is suitable as a capture antibody for secukinumab in a pharmacokinetic (PK) bridging ELISA to measure free drug when paired with either clone AbD37054ia (HCA371) or clone AbD37091ia (HCA372), in IgG1 format, as the detection reagent. Secukinumab (Cosentyx) is a human IgG1/kappa antibody generated using transgenic mouse technology. It has been approved for the treatment of plaque psoriasis, ankylosing spondylitis, psoriatic arthritis and non-radiographic axial spondyloarthritis. This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.
