A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, the hIL-17A/secukinumab complex was formed by adding 5 µg/ml secukinumab to the wells coated with hIL-17A. Detection was performed using Human Anti-Secukinumab (Drug/Target Complex) Antibody, clone AbD36949 (HCA375) at a concentration of 2 μg/ml followed by an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.

After washing and blocking with PBST + 5% BSA, the hIL-17A/secukinumab complex was formed by adding 2 µg/ml secukinumab to the wells coated with hIL-17A. Detection was performed using Human Anti-Secukinumab (Drug/Target Complex) Antibody, clone AbD36949 (HCA375) titrated to the given concentrations in PBST followed by Mouse Anti-Penta Histidine Tag:HRP Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

Data are shown as the mean of three measurements.

| Human Anti-Secukinumab (Drug/Target Complex) Antibody, clone AbD36949 specifically recognizes the human monoclonal antibody drug secukinumab when in complex with its target, interleukin 17A (IL-17A). The antibody does not recognize free secukinumab or unbound IL-17A. It can be used to measure levels of secukinumab or its biosimilars in patient samples. Clone AbD36949 is a monovalent Fab format antibody that can be used to develop a pharmacokinetic (PK) antigen capture assay to measure free secukinumab that has been captured via immobilized IL-17A. Secukinumab (Cosentyx) is a human IgG1/kappa antibody generated using transgenic mouse technology. It has been approved for the treatment of plaque psoriasis, ankylosing spondylitis, psoriatic arthritis and non-radiographic axial spondyloarthritis.
