A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Secukinumab Antibody, clone AbD36536ia (HCA373) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Secukinumab Antibody, clone AbD36536ia (HCA373) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Secukinumab Antibody, clone AbD37054ia (HCA371, black), clone AbD37091ia (HCA372, red) or clone AbD36536ia (HCA373, blue). Detection was performed using HRP conjugated secukinumab at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human IL-17A at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of secukinumab (0.3 µg/ml) plus increasing concentrations of Human Anti-Secukinumab Antibody, clone AbD36536ia (HCA373) in the monovalent Fab format was added. Free secukinumab, still capable of binding to the human IL-17A coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Secukinumab Antibody, clone AbD36536ia is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the human monoclonal antibody secukinumab. It does not recognize free human interleukin IL-17A (IL-17A) nor secukinumab in complex with human IL-17A. The antibody can be used to measure secukinumab or its biosimilars in patient samples. Clone AbD36536ia is a fully human recombinant monoclonal antibody with IgG1 isotype and can be used as a surrogate positive control or reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 12 nM, and is one of three antibodies with different affinities offered for ADA assays. Secukinumab (Cosentyx) is a human IgG1/kappa antibody generated using transgenic mouse technology. It has been approved for the treatment of plaque psoriasis, ankylosing spondylitis, psoriatic arthritis and non-radiographic axial spondyloarthritis. This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.
