Pertuzumab antibody | AbD35668ia
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Pertuzumab Antibody, clone AbD35668ia (HCA344) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with human HER2, pertuzumab, trastuzumab, human IgG1 kappa or lambda at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the human HER2/pertuzumab complex was formed by adding 5 µg/ml pertuzumab to the wells coated with human HER2, detection was performed using HRP conjugated Human Anti-Pertuzumab Antibody, clone AbD35668ia (HCA344) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for Pertuzumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Pertuzumab Antibody, clone AbD35668ia (HCA344, black), or clone AbD37069ia (HCA345, red). Detection was performed using HRP conjugated pertuzumab followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with HER2 at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of pertuzumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Pertuzumab Antibody, clone AbD35668ia (HCA344) in the monovalent Fab format (the same as clone AbD35668, HCA341) was added. Free pertuzumab, still capable of binding to the HER2 coated plate, was detected using an HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Pertuzumab Antibody, clone AbD35668ia is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the humanized monoclonal antibody pertuzumab. It does not recognize free human epidermal growth factor receptor 2 (HER2). The antibody can be used to measure free pertuzumab or its biosimilars in patient samples. Clone AbD35668ia is a fully human recombinant monoclonal antibody with IgG1 isotype and can be used as a surrogate positive control or reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 14 nM, and is one of two antibodies with different affinities offered for ADA assays. Pertuzumab (Perjeta) is an IgG1/kappa antibody humanized from mouse. It has been approved for the treatment of patients with metastatic HER2-positive breast cancer in combination with trastuzumab. Pertuzumab binds to the extracellular domain II of HER2 and inhibits HER2–HER3 dimerization, reducing signaling through intracellular pathways (Capelan et al. 2013). This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.
