A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Pertuzumab (Drug/Target Complex) Antibody, clone AbD35572ad (TZA035) at a concentration of 2 μg/ml followed by a HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with human HER2, pertuzumab, trastuzumab or human IgG1 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the human HER2/Pertuzumab complex was formed by adding 5 µg/ml pertuzumab to the wells coated with human HER2. Detection was performed using Human Anti-Pertuzumab (Drug/Target Complex) Antibody, clone AbD35572ad (TZA035) titrated to the given concentrations in PBST followed by a HRP conjugated Anti-Penta His tag Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for human HER2/Pertuzumab is shown as the mean of three measurements.

Detection was performed using HRP conjugated Human Anti-Pertuzumab (Drug/Target Complex) Antibody, clone AbD35572pap (TZA035P) at a concentration of 0.5 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Pertuzumab (Drug/Target Complex) Antibody, clone AbD35572 specifically recognizes the antibody drug pertuzumab when in complex with its target, the human epidermal growth factor receptor 2 (HER2). It does not recognize free pertuzumab nor unbound HER2. The monovalent Fab format of this antibody, clone AbD35572ad (TZA035) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is also available as a bivalent Fab antibody pre-coupled to the HRP-conjugated BiSpyCatcher2 (TZC002P) under product code TZA035P for direct detection of the pertuzumab/HER2 complex in bioanalysis assays. Clone AbD35572 can be used in pharmacokinetic (PK) antigen capture format assays to measure pertuzumab that has be captured via immobilized HER2. It is ideal for bioanalytical assays to measure the level of drug or biosimilar in complex with the HER2 target. Pertuzumab (Perjeta) is an IgG1/kappa antibody humanized from mouse. It has been approved for the treatment of patients with metastatic HER2-positive breast cancer in combination with trastuzumab. Pertuzumab binds to the extracellular domain II of HER2 and inhibits HER2–HER3 dimerization, reducing signaling through intracellular pathways (Capelan et al. 2013).
