A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, the dupilumab/IL-4Rα complex was formed by adding dupilumab at the concentration of 5 μg/ml to wells coated with IL-4Rα. Detection was performed using Human Anti-Dupilumab (Drug/Target complex) Antibody, clone AbD48337ad (TZA054) at a concentration of 2 μg/ml, followed by a an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.

After washing and blocking with PBST + 5% BSA, the dupilumab/IL-4Rα complex was formed by adding 5 µg/ml dupilumab to the wells coated with IL-4Rα. Detection was performed using Human Anti-Dupilumab (Drug/Target complex) Antibody, clone AbD48337ad (TZA054) titrated to the given concentrations in PBST, followed by a an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of dupilumab. Detection was performed using Human Anti-Dupilumab (Drug/Target complex) Antibody, clone AbD48337ad (TZA054) at a concentration of 0.5 mg/ml in PBST, followed by a an HRP conjugated anti-DYKDDDDK tag antibodyin HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Dupilumab (Drug/Target Complex) Antibody, clone AbD48337 recognizes the monoclonal antibody drug dupilumab when in complex with its target, the Interleukin-4 Receptor alpha (IL-4Rα). It does not recognize free dupilumab or unbound IL-4Rα. It can be used to measure levels of dupilumab in patient samples. Clone AbD48337 is a monovalent Fab format antibody that can be used to develop a pharmacokinetic (PK) antigen capture assay to measure free dupilumab captured via immobilized IL-4Rα. It is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour.
