A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Burosumab Antibody, clone AbD63870ia (HCA460) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with burosumab, recombinant His-tagged human FGF-23, Adalimumab, human IgG1 kappa control and human IgG1 lambda control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Burosumab Antibody, clone AbD63870ia (HCA460) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for burosumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Burosumab Antibody, clone AbD63865ia (HCA459, black), clone AbD63872ia (HCA461, blue), or clone AbD63870ia (HCA460, red). Detection was performed using HRP conjugated burosumab at a concentration of 2μg/ml in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of burosumab (0.15 µg/mL) plus increasing concentrations of Human Anti-Burosumab Antibody, clone AbD63870 (HCA460) in the monovalent Fab format was added. Free burosumab, still capable of binding to the human FGF-23-His coated plate, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. As a negative control, Human anti-Burosumab, clone AbD64797ad (TZA0111) was also tested.

E Reset| Human Anti-Burosumab Antibody, clone AbD63870ia is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug burosumab. It does not recognize the drug target hFGF-23-His, nor burosumab in complex with hFGF-23-His. HCA460 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 0.6nM and is one of the three antibodies offered for ADA assays. Antibody HCA459 has an affinity of 0.1nM and HCA461 has an affinity of 6.2nM. Burosumab (trade name Crysvita) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds to fibroblast growth factor-23 (FGF23), blocking the interaction with the FGFR1. Burosumab therefore inhibits FGF23 signaling, thereby increasing tubular phosphate reabsorption and decreasing renal phosphate excretion, as well as increasing serum levels of 1,25(OH)2D and increasing GI absorption of phosphate. As a result, bone mineralization is improved and the risk of bone disease is decreased. Burosumab is indicated for the treatment of X-linked hypophosphatemia and tumor-induced osteomalacia.
