Human Anti-Burosumab (Drug/Target Complex) Antibody, clone AbD64797 specifically recognizes the antibody drug burosumab when in complex with its target, hFGF-23-His. It does not recognize free burosumab nor unbound hFGF-23-His.

The monovalent Fab format of this antibody, clone AbD64797ad (TZA0111) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour.
This antibody is also available as a bivalent Fab antibody pre-coupled to the HRP-conjugated BiSpyCatcher2 (TZC002P) under product code TZA0111P for direct detection of the Burosumab/ hFGF-23-His complex in bioanalysis assays.

Clone AbD64797 can be used in pharmacokinetic (PK) antigen capture format assays to measure burosumab that has been captured via immobilized hFGF-23-His. It is ideal for bioanalytical assays to measure the level of drug or biosimilar in complex with the hFGF-23-His target.

Burosumab (trade name Crysvita) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds to fibroblast growth factor-23 (FGF23), blocking the interaction with the FGFR1. Burosumab therefore inhibits FGF23 signaling, thereby increasing tubular phosphate reabsorption and decreasing renal phosphate excretion, as well as increasing serum levels of 1,25(OH)2D and increasing GI absorption of phosphate. As a result, bone mineralization is improved and the risk of bone disease is decreased. Burosumab is indicated for the treatment of X-linked hypophosphatemia and tumor-induced osteomalacia.

Product Form
A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA0111 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
