Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Romosozumab Antibody, clone AbD60146ad (TZA061) at a concentration of 2 μg/mL and an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with romosozumab, Sclerostin-His, human IgG2 kappa control and normal human serum (NHS) at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Romosozumab Antibody, clone AbD60146ad (TZA061) titrated to the given concentrations followed by an HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for romosozumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of romosozumab. Detection was performed using HRP-conjugated Human Anti-Romosozumab Antibody, clone AbD60146pap (TZA061P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of romosozumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Romosozumab Antibody, clone AbD60146ad (TZA061). Free romosozumab, still capable of binding to the Sclerostin-His coated plate, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Romosozumab Antibody, clone AbD60146 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug romosozumab, and its biosimilar products. It does not recognize the drug target, sclerostin, nor romosozumab in complex with sclerostin. This antibody can be used to measure free romosozumab levels in serum from patients. Clone AbD60146 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA061. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA061P. A pair of anti-romosozumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD60146 is recommended as a detection antibody, paired with Human Anti-Romosozumab Antibody, clone AbD60141ad (TZA060) as the capture reagent. Romosozumab (trade name Evenity) is a humanized monoclonal antibody of the IgG2/kappa isotype that binds to sclerostin and prevents inhibition of the Wnt signaling pathway, thereby promoting osteogenesis and, to a lesser extent, reducing bone resorption. Romosozumab's inhibition of sclerostin also inhibits the increase in RANKL-dependent increases in osteoclast activity and bone resorption. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab TZA061 with a HRP conjugated BiSpyCatcher version 2. The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
