E Reset| Human Anti-Romosozumab Antibody, clone AbD60141ad is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug romosozumab, and its biosimilar products. It does not recognize the drug target, sclerostin, nor romosozumab in complex with sclerostin. This antibody can be used to measure free romosozumab levels in serum from patients. A pair of anti-romosozumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD60141ad is recommended as a capture antibody, paired with Human Anti-Romosozumab Antibody, clone AbD60146 (TZA061) as the detection antibody. TZA060 is a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog. Romosozumab (trade name Evenity) is a humanized monoclonal antibody of the IgG2/kappa isotype that binds to sclerostin and prevents inhibition of the Wnt signaling pathway, thereby promoting osteogenesis and, to a lesser extent, reducing bone resorption. Romosozumab's inhibition of sclerostin also inhibits the increase in RANKL-dependent increases in osteoclast activity and bone resorption. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.
