A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL, including Efgartigimod, Nirsevimab and AbD18705iu, which carry the YTE mutations. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-YTE Mutations Antibody, clone AbD64789ad (TZA0136) at a concentration of 2 μg/mL followed by HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with AbD18705iu carrying the YTE mutations, as well as human IgG1, human IgG4 pro control, and cyno IgG as controls without the YTE mutations at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-YTE Mutations Antibody, clone AbD64789pap (TZA0136P) titrated to the given concentrations in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for AbD18705iu are shown as the mean of two measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of Recombinant Human IgG1 YTE Kappa Allotype G1m3 ((HCA413) carrying the YTE-mutations. Detection was performed using HRP-conjugated Human Anti-YTE mutations Antibody, clone AbD64789pap (TZA0136P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-YTE mutation Antibody, clone AbD64789 is an antibody that specifically recognizes the YTE mutation (M252Y/S254T/T256E) in human IgG1 antibodies. This antibody can be used to measure antibody drug concentrations in serum from patients treated with antibody drugs carrying this YTE mutation. For PK assays, clone AbD64789 is recommended as a detection antibody, paired with Human anti-YTE mutation antibody, clone AbD64785ad (TZA0135) as the capture antibody. Clone AbD64789 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0136. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0136P. The YTE-mutations (M252Y/S254T/T256E) were initially introduced at the CH2-CH3 interface of the humanized IgG1 drug antibody MEDI-524 (anti-respiratory syncytial virus, RSV). It was shown that these mutations lead to a prolonged half time in serum due to a higher affinity to the neonatal Fc receptor (FcRn) and is therefore an interesting format for many antibody drugs. - A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA0136 with a HRP conjugated BiSpyCatcher version 2. The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.

- ELISA
- Clone AbD64789pap can be used in indirect ELISA for AbD18705iu and other antibodies carrying the YTE mutations. It can also be used as a detection antibody to develop a pharmacokinetic (PK) bridging ELISA together with TZA0135 as the capture reagent. Note: This antibody has only low affinity towards YTE in the presence of additional Fc mutations. For antibodies carrying additional mutations we recommend to use our alternative PK pair of TZA0137 and TZA0137P.
