Human Anti-Ixekizumab Antibody, clone AbD60554 is a non-paratope specific, non-inhibitory, anti-idiotypic antibody (Type 2) that specifically recognizes the free, partially bound and fully bound human monoclonal antibody drug ixekizumab, and its biosimilar products. This antibody can be used to measure free, partially bound and fully bound ixekizumab levels in serum from patients.

This TrailBlazer Antibody is available in an unconjugated monovalent Fab format. It may be used as capture antibody for ixekizumab in a pharmacokinetic (PK) antigen detection assay together with the bivalent Fab conjugated to HRP, TZA0107P (AbD48542ad) as detection reagent.

Ixekizumab (trade name Taltz) is a humanized monoclonal antibody of the IgG4/kappa isotype that binds to interleukin 17 (IL-17A), a pro-inflammatory cytokine, and blocks its action. Blocking its effect is beneficial for use in inflammatory conditions. Ixekizumab is indicated for the treatment of moderate-to-severe plaque psoriasis and as a second-line therapy for active psoriatic arthritis.

Product Form
A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL phage display library and expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.

This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.

Avoid repeated freezing and thawing as this may denature the antibody. Storage in frost-free freezers is not recommended.

Clone AbD60554ad can be used in indirect ELISA to detect ixekizumab. It can also be used as a capture antibody to develop a pharmacokinetic (PK) bridging ELISA for ixekizumab, together with TZA0107P as the detection reagent.
