Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ixekizumab Antibody, clone AbD60554ad (TZA0107) at a concentration of 2 μg/ml followed by HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with ixekizumab, nivolumab, IgG4 isotype control, human IgG4 kappa control and human IgG4 lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ixekizumab Antibody, clone AbD60554ad (TZA0107) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for ixekizumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of ixekizumab. Detection was performed using HRP conjugated Human Anti-Ixekizumab Antibody, clone AbD60554pap (TZA0107P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human IL-17A at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of ixekizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Ixekizumab Antibody, clone AbD60554ad (TZA0107) was added. The inhibitory Human Anti-Ixekizumab Antibody, clone AbD60570ad (TZA0105) was used as a negative control. Free ixekizumab, still capable of binding to the human IL-17A coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Ixekizumab Antibody, clone AbD60554 is a non-paratope specific, non-inhibitory, anti-idiotypic antibody (Type 2) that specifically recognizes the free, partially bound and fully bound human monoclonal antibody drug ixekizumab, and its biosimilar products. This antibody can be used to measure free, partially bound and fully bound ixekizumab levels in serum from patients. This TrailBlazer Antibody is available in an unconjugated monovalent Fab format. It may be used as capture antibody for ixekizumab in a pharmacokinetic (PK) antigen detection assay together with the bivalent Fab conjugated to HRP, TZA0107P (AbD48542ad) as detection reagent. Ixekizumab (trade name Taltz) is a humanized monoclonal antibody of the IgG4/kappa isotype that binds to interleukin 17 (IL-17A), a pro-inflammatory cytokine, and blocks its action. Blocking its effect is beneficial for use in inflammatory conditions. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL phage display library and expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.
