A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Nirsevimab Antibody, clone AbD64625ia (HCA451) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with nirsevimab, human IgG1 kappa control, VH/VL subclass control and normal human serum (NHS) at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Nirsevimab Antibody, clone AbD64625ia (HCA451) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for nirsevimab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Nirsevimab Antibody, clone AbD64623ia (HCA450, black), clone AbD64626ia (HCA452, blue), or clone AbD64625ia (HCA451, red). Detection was performed using biotin conjugated Nirsevimab at a concentration of 2 µg/mL in PBST followed by HRP conjugated Neutravidin in HISPEC Assay Diluent (BUF049A). Biotin conjugation of the antibody used in this assay was performed using a LYNX Rapid Plus Biotin (Type 1) Antibody Conjugation Kit (LNK263B). Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with fusion glycoprotein F0 of RSV at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, pre-incubated mixture of nirsevimab (0.15 µg/mL) plus increasing concentrations of Human Anti-Nirsevimab Antibody, clone AbD64625ia (HCA451) in the monovalent Fab format was added. Free nirsevimab, still capable of binding to the fusion glycoprotein F0 of RSV coated plate, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Nirsevimab Antibody, clone AbD64625ia is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug nirsevimab, and its biosimilar products. It does not recognize the free Fusion glycoprotein F0 of RSV, nor nirsevimab in complex with Fusion glycoprotein F0 of RSV. HCA451 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 0.5nM and is one of the three antibodies offered for ADA assays. Antibody HCA450 has an affinity or 0.1nM and HCA452 has an affinity of 2.7nM. Nirsevimab (trade name Beyfortus) is a humanized monoclonal antibody of the IgG1/kappa isotype that targets the prefusion conformation of the RSV F protein, a glycoprotein involved in the membrane fusion step of the viral entry process and neutralises several RSV A and B strains. Nirsevimab is indicated to treat respiratory syncytial virus (RSV) lower respiratory tract disease in neonates and infants. |
