A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Pertuzumab Antibody, clone AbD35569ad (TZA025) at a concentration of 2 μg/ml followed by a HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with human HER2, pertuzumab, trastuzumab or human IgG1 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the human HER2/Pertuzumab complex was formed by adding 5 µg/ml pertuzumab to the wells coated with human HER2. Detection was performed using Human Anti-Pertuzumab Antibody, clone AbD35569ad (TZA025) titrated to the given concentrations in PBST followed by a HRP conjugated Anti-Penta His tag Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for pertuzumab and human HER2/Pertuzumab is shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of pertuzumab. Detection was performed using HRP conjugated Human Anti-Pertuzumab Antibody, clone AbD35569pap (TZA025P) at a concentration of 0.5 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Pertuzumab Antibody, clone AbD35569 is a non-paratope specific, non-inhibitory anti-idiotypic antibody that specifically recognizes the humanized monoclonal antibody drug pertuzumab. It recognizes free pertuzumab and pertuzumab in complex with its target, the human epidermal growth factor receptor 2 (HER2) but not free HER2. It can be used to measure total levels of pertuzumab and biosimilars in patient samples. The monovalent Fab format of this antibody, clone AbD35569ad (TZA025) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is also available as a bivalent Fab antibody pre-coupled to the HRP-conjugated BiSpyCatcher2 (TZC002P) under product code TZA025P for direct detection of pertuzumab in bioanalysis assays. A pair of anti-pertuzumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure total drug. Clone AbD35569 can be used as a detection antibody to develop a pharmacokinetic (PK) bridging assay together with HCA341 as the capture antibody. Pertuzumab (Perjeta) is an IgG1/kappa antibody humanized from mouse. It has been approved for the treatment of patients with metastatic HER2-positive breast cancer in combination with trastuzumab. Pertuzumab binds to the extracellular domain II of HER2 and inhibits HER2–HER3 dimerization, reducing signaling through intracellular pathways (Capelan et al. 2013).
