Panitumumab antibody | AbD23897_hIgG1
A microtiter plate was coated over night with various antigens at a concentration of 1 μg/ml. After washing and blocking with PBST+5% BSA, detection was performed using HRP-conjugated Human anti panitumumab (clone AbD23897_hIgG1) at a concentration of 2 μg/ml in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.

After washing and blocking with PBST+5% BSA, detection was performed using HRP conjugated Human anti panitumumab (clone AbD23897_hIgG1) at a concentration of 2 μg/ml in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for binding to panitumumab is shown as the mean of three measurements.

After washing and blocking with PBST+5% BSA, 10% human serum was added spiked with increasing concentrations of panitumumab. Detection was performed using HRP conjugated Human anti panitumumab (clone AbD23897_hIgG1, HCA265P) at a concentration of 2 μg/ml in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.

After washing and blocking with PBST+5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of human anti panitumumab HCA265 (clone AbD23897_hIgG1) or HCA266 (clone AbD23895_hIgG1). Detection was performed using HRP conjugated panitumumab. Data is shown as the mean of three measurements.
A microtiter plate was coated over night with human EGFR at a concentration of 1 μg/ml. After washing and blocking with PBST+5% BSA, a pre-incubated mixture of panitumumab (0.3 μg/ml) plus increasing concentrations of Human anti panitumumab (clone AbD23897) in the monovalent Fab format was added. Free panitumumab, still capable of binding to the EGFR-coated plate, was detected using Goat anti Human IgG F(ab')2 HRP (0500-0099) and QuantaBlu fluorogenic peroxidase substrate. Data is shown as the mean of three measurements.

| Human anti panitumumab, clone AbD23897_hIgG1, is a paratope specific, high affinity, anti-idiotypic antibody that specifically recognizes the fully human monoclonal antibody drug panitumumab and inhibits it binding to its target, epidermal growth factor receptor (EGFR). The antibody can be used to measure the levels of panitumumab and biosimilar products in bioanalytical assays. Clone AbD23897_hIgG1 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a reference standard in an anti-drug antibody (ADA) assay. Additionally the antibody can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. This antibody, in full immunoglobulin format, is recommended as the detection antibody, paired with the antibody clone AbD23894 (HCA264) in monovalent Fab format, as the capture antibody. Panitumumab (Vectibix®) is a fully human monoclonal antibody (IgG2/kappa) specific to the transmembrane protein EGFR. The drug binds to the extracellular domain of the EGFR, inhibiting the activation of the intracellular signaling cascade.
