Ocrelizumab antibody | AbD50651
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ocrelizumab Antibody, clone AbD50651ad (TZA045) at a concentration of 2 μg/ml followed by an HRP-labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with ocrelizumab, rituximab, infliximab, human IgG1 kappa control (same allotype as ocrelizumab and rituximab) or human lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ocrelizumab Antibody, clone AbD50651ad (TZA045) titrated to the given concentrations in PBST followed by an HRP-labelled anti-DYKDDDDK tag antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

All experiments were performed on a Daudi cell line expressing CD20 and incubated for 1 hour with the antibody mix.

Data are shown as the mean of three measurements.

| Human Anti-Ocrelizumab Antibody, clone AbD50651 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug ocrelizumab and its biosimilar products. Consequently, this antibody can be used to measure free ocrelizumab levels in serum from patients. It does not cross react with rituximab. This TrailBlazer Antibody is available in a purified monovalent Fab format tagged with a SpyTag2 at the C terminus of the Fab antibody heavy chain (TZA044), enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. It is also available as a bivalent Fab format pre-coupled to an HRP-conjugated BiSpyCatcher2 (TZC002P) for direct detection of ocrelizumab under product code TZA045P. Ocrelizumab (trade name Ocrevus) is a monoclonal IgG1/kappa antibody (VH3/κ1). In vitro, it binds to CD20 cell surface molecules on target cells thereby inducing immune effector mechanisms such as antibody-dependent cellular phagocytosis (ADCP), antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and apoptosis. In vivo, ocrelizumab selectively and effectively depletes CD20+ B cells. Hence, it is an immunosuppressive drug.
