Ocrelizumab antibody | AbD50641ad
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ocrelizumab Antibody, clone AbD50641ad (TZA044) at a concentration of 2 μg/ml followed by an HRP-labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with ocrelizumab, rituximab, infliximab, human IgG1 kappa control (same allotype as ocrelizumab and rituximab) or human lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ocrelizumab Antibody, clone AbD50641ad (TZA044) titrated to the given concentrations in PBST followed by an HRP-labelled anti-DYKDDDDK tag antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

All experiments were performed on a Daudi cell line expressing CD20 and incubated for 1 hour with the antibody mix.

| Human Anti-Ocrelizumab Antibody, clone AbD50641ad is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug ocrelizumab and its biosimilar products. Consequently, this antibody can be used to measure free ocrelizumab levels in serum from patients. This antibody is available in two formats - a monovalent Fab unconjugated format, clone AbD50641ad (TZA044), which has a SpyTag incorporated. This format can be used as a capture antibody in a pharmacokinetic (PK) bridging ELISA together with AbD50651 as a detection antibody. Clone AbD50641 is also available in full length IgG1 format under product code HCA400 which can be used in an ADA bridging ELISA as a surrogate positive control. Human Anti-Ocrelizumab Antibody, clone AbD50641ad has been shown to cross-react with the antibody drug rituximab (tradename Rituxan). Ocrelizumab is the humanized form of rituximab. There are changes in at least one of the CDRs and it contains Fc modification. Ocrelizumab (trade name Ocrevus) is a monoclonal IgG1/kappa antibody (VH3/κ1). In vitro, it binds to CD20 cell surface molecules on target cells thereby inducing immune effector mechanisms such as antibody-dependent cellular phagocytosis (ADCP), antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and apoptosis. In vivo, ocrelizumab selectively and effectively depletes CD20+ B cells. Hence, it is an immunosuppressive drug.
