Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ixekizumab Antibody, clone AbD60557ad (TZA0106) at a concentration of 2 μg/ml followed by HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with ixekizumab, nivolumab, IgG4 isotype control, human IgG4 kappa control and human IgG4 lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Ixekizumab Antibody, clone AbD60557ad (TZA0106) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for ixekizumab are shown as the mean of three measurements.
A microtiter plate was coated overnight with Human Anti-Ixekizumab Antibody, clone AbD60570ad (TZA0105) at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of ixekizumab. Detection was performed using HRP conjugated Human Anti-Ixekizumab Antibody, clone AbD60557pap (TZA0106P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human IL-17A at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of ixekizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Ixekizumab Antibody, clone AbD60557ad (TZA0106) was added. Free ixekizumab, still capable of binding to the human IL-17A coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. The non-inhibitory Human Anti-Ixekizumab Antibody, clone AbD60554ad (TZA0107) was used as a negative control. Data are shown as the mean of three measurements.

E Reset| Human Anti-Ixekizumab Antibody, clone AbD60557 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug ixekizumab, and its biosimilar products. It does not recognize the drug target, human Interleukin 17 (IL-17A), nor ixekizumab in complex with IL-17A. This antibody can be used to measure free ixekizumab levels in serum from patients. Clone AbD60557 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0106. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0106P. A pair of anti-ixekizumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD60557 is recommended as a detection antibody, paired with Human Anti-Ixekizumab Antibody, clone AbD60570ad (TZA0105) as the capture reagent. Ixekizumab (trade name Taltz) is a humanized monoclonal antibody of the IgG4/kappa isotype that binds to interleukin 17 (IL-17A), a pro-inflammatory cytokine, and blocks its action. Blocking its effect is beneficial for use in inflammatory conditions. - A monovalent human recombinant Fab (kappa light chain) selected from the HuCAL phage display library and expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.

- ELISA
- Clone AbD60557ad can be used in indirect ELISA to detect ixekizumab. It can also be used as a detection antibody to develop a pharmacokinetic (PK) bridging ELISA for risankizumab, together with TZA0105 as the capture reagent. Note that both TZA0105 (capture) and TZA0106 (detection) contain the same peptide tags. Therefore, using anti-tag antibodies for detecting TZA0106 is not possible.
