A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Dulaglutide Antibody, clone AbD64818ia (HCA449) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with dulaglutide, GLP-1 TRF, human IgG4-pro control, human IgG4-LALA control, and galcanezumab at a concentration of 5μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti dulaglutide antibody, clone AbD64818ia (HCA449) titrated to given concentrations in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with dulaglutide-Fc at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Dulaglutide Antibody, clone AbD64818ia (HCA449). Detection was performed using biotinylated dulaglutide at a concentration of 2 µg/mL in HISPEC Assay Diluent (BUF049A) followed by HRP conjugated Neutravidin and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Dulaglutide Antibody, clone AbD64818ia is specific for human GLP-1 and its peptidase-IV-protected analog present in the human IgG4-Fc fusion protein dulaglutide. HCA449 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 2.0nM. Dulaglutide (trade name Trulicity) is a fusion protein, combining a modified GLP-1 analogue with a human IgG4-Fc heavy chain. It binds to GLP-1 and activates the GLP-1 receptor found in the pancreatic beta cells increasing intracellular cyclic AMP in beta cells, leading to insulin release and subsequent reduction of blood glucose concentrations.
