Dulaglutide (Trulicity) is a GLP‑1 receptor agonist made of a GLP‑1 peptide fused to a human IgG4‑Fc fragment. The Fc part has the IGHG4*04 allotype containing a LALA mutation. Dulaglutide activates the GLP-1 receptor found in pancreatic beta cells, increasing intracellular cyclic adenosine monophosphate (cAMP) and stimulating insulin release, subsequently reducing blood glucose concentration. Additionally, dulaglutide decreases glucagon secretion and slows gastric emptying.
Anti-dulaglutide antibodies, clones AbD64818ad and AbD64818ia, are recombinant antibodies that are specific for GLP-1 and recognize the free GLP-1/IgG4-Fc fusion protein, dulaglutide. These clones also recognize human GLP-1 coupled to carrier proteins.
They are ideal for the development of a pharmacokinetic (PK) bridging ELISA to measure free drug. In a fully human IgG1 format, they are suitable as a surrogate positive control or calibrator for an anti-drug antibody (ADA) assay.
