A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL, including the GLP-1 BSA complex, GLP-1 transferrin complex and GLP-1 IgG4-Fc fusion protein dulaglutide. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Dulaglutide Antibody, clone AbD64818pap (TZA0123P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with dulaglutide, human GLP-1 coupled to Transferrin, human IgG4-pro isotype control, human IgG4-LALA isotype control, and galcanezumab at a concentration of 5 µg/mL. After washing and blocking with PBS + 5% BSA, detection was performed using Human Anti-Dulaglutide Antibody, clone AbD64818ad (TZA0123) titrated to the given concentrations followed by an HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for dulaglutide are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of dulaglutide. Detection was performed using HRP-conjugated Human Anti-Dulaglutide Antibody, clone AbD64818pap (TZA0123P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Dulaglutide Antibody, clone AbD64818 is specific for human GLP-1 and its peptidase-IV-protected analog present in the human IgG4-Fc fusion protein dulaglutide. Clone AbD64818 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0123. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0123P. A pair of anti-dulaglutide antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD64818ad is recommended as a capture antibody, paired with Human Anti-Dulaglutide Antibody, clone AbD64818pap (TZA0123P) as the detection reagent. Dulaglutide (trade name Trulicity) is a fusion protein, combining a modified GLP-1 analogue with a human IgG4-Fc heavy chain. It binds to GLP-1 and activates the GLP-1 receptor found in the pancreatic beta cells increasing intracellular cyclic AMP in beta cells, leading to insulin release and subsequent reduction of blood glucose concentrations.
