Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-hFGF-23-His/Burosumab Antibody, clone AbD64797ad (TZA0111) at a concentration of 2 μg/ml in PBST followed by an HRP labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with hFGF-23-His, burosumab, human IgG1 kappa control and human IgG1 lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the hFGF-23-His/burosumab complex was formed by adding 5µg/mL burosumab to the wells coated with hFGF-23-His. Detection was performed using Human Anti-Burosumab (Drug/Target Complex) Antibody, clone AbD64797ad (TZA0111) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for burosumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of burosumab. Detection was performed using Human Anti-Burosumab (Drug/Target Complex) Antibody, clone AbD64797pap (TZA0111P) at a concentration of 0.01 µg/ml in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Burosumab (Drug/Target Complex) Antibody, clone AbD64797 specifically recognizes the antibody drug burosumab when in complex with its target, hFGF-23-His. It does not recognize free burosumab nor unbound hFGF-23-His. The monovalent Fab format of this antibody, clone AbD64797ad (TZA0111) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is also available as a bivalent Fab antibody pre-coupled to the HRP-conjugated BiSpyCatcher2 (TZC002P) under product code TZA0111P for direct detection of the Burosumab/ hFGF-23-His complex in bioanalysis assays. Clone AbD64797 can be used in pharmacokinetic (PK) antigen capture format assays to measure burosumab that has been captured via immobilized hFGF-23-His. It is ideal for bioanalytical assays to measure the level of drug or biosimilar in complex with the hFGF-23-His target. Burosumab (trade name Crysvita) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds to fibroblast growth factor-23 (FGF23), blocking the interaction with the FGFR1. Burosumab therefore inhibits FGF23 signaling, thereby increasing tubular phosphate reabsorption and decreasing renal phosphate excretion, as well as increasing serum levels of 1,25(OH)2D and increasing GI absorption of phosphate. As a result, bone mineralization is improved and the risk of bone disease is decreased. Burosumab is indicated for the treatment of X-linked hypophosphatemia and tumor-induced osteomalacia. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.
