Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Burosumab Antibody, clone AbD63865ad (TZA0110) at a concentration of 2 μg/ml in PBST followed by an HRP labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with Burosumab, recombinant His-tagged human FGF-23, Adalimumab, human IgG1 kappa control and human IgG1 lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Burosumab Antibody, clone AbD63865ad (TZA0110) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for burosumab are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant His-tagged human FGF-23 at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of burosumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Burosumab Antibody, clone AbD63865ad (TZA0110) was added. Free burosumab, still capable of binding to the human FGF-23-His coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. The complex-specific Human Anti-hFGF-23-His/Burosumab Antibody (Type 3), clone AbD64797ad (TZA0111) was used as a negative control. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of burosumab. Detection was performed using HRP conjugated Human Anti-Burosumab Antibody, clone AbD63865pap (TZA110P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti- Burosumab Antibody, clone AbD63865 is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the humanized monoclonal antibody drug burosumab. It does not recognize the drug target hFGF-23-His, nor burosumab in complex with hFGF-23-His. This antibody can be used to measure free burosumab levels in serum from patients. TZA0110 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0110. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0110P. A pair of anti-burosumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD63865ad is recommended as a detection antibody, paired with Human Anti-Burosumab Antibody, clone AbD63869ad (TZA0109) as the capture antibody. Burosumab (trade name Crysvita) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds to fibroblast growth factor-23 (FGF23), blocking the interaction with the FGFR1. Burosumab therefore inhibits FGF23 signaling, thereby increasing tubular phosphate reabsorption and decreasing renal phosphate excretion, as well as increasing serum levels of 1,25(OH)2D and increasing GI absorption of phosphate. As a result, bone mineralization is improved and the risk of bone disease is decreased. Burosumab is indicated for the treatment of X-linked hypophosphatemia and tumor-induced osteomalacia.
