Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Benralizumab Antibody, clone AbD60166ia (HCA428) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Benralizumab Antibody, clone AbD60166ia (HCA428) titrated to the given concentrations in PBST followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for benralizumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Benralizumab Antibody, clone AbD60095ia (HCA426, black), clone AbD59975ia (HCA427, blue) or clone AbD60166ia (HCA428, red). Detection was performed using HRP conjugated canakinumab at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of benralizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Benralizumab Antibody, clone AbD60166ia (HCA428) in the monovalent Fab format was added. Free Benralizumab, still capable of binding to the IL5Ra-His coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Benralizumab Antibody, clone AbD60166ia is a paratope specific, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug benralizumab. It does not recognize the drug target, human interleukin 5 receptor α (IL5Rα), nor benralizumab in complex with IL5Rα.. HCA428 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 6nM and is one of the three antibodies offered for ADA assays. Antibody HCA426 has an affinity of 0.2nM and HCA427 has an affinity of 4nM. Benralizumab (trade name Fasenra) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds with high affinity to a conformationally distinct epitope within domain 1 of the α-chain of IL-5R, and thus inhibits proliferation of IL-5-dependent cell lines, and otherwise blocks signaling through the IL-5 receptor.
